华中科大史岸冰课题组报道WTS-1/LATS调控内吞循环运输的新机制

2021年11月24日,华中科技大学基础医学院史岸冰课题组Journal of Cell Science (JCS) 在线发表了题为“WTS-1/LATS regulates endocytic recycling by restraining F-actin assembly in a synergistic manner”的研究论文,报道了激酶WTS-1/LATS以协同方式限制内吞体上微丝结构组装以调控内吞循环运输。


华中科技大学基础医学院博士研究生张晗翀程子航为该论文的共同第一作者,史岸冰教授林珑副教授为共同通讯作者。

细胞外大分子,配体/受体复合物、功能膜蛋白、膜脂等通过内吞作用内化进入细胞,随后在胞内囊泡状或管状膜性内吞体结构中进行一系列的分选和定向运输[1]。大分子经过分选后,一部分由内吞循环运输回到质膜,一部分通过逆向运输被送到高尔基体,还有一部分被送到溶酶体降解[2]。对于功能膜蛋白和膜脂,内吞的持续发生会导致其细胞膜丰度的降低,需要通过循环运输将大分子物质送回质膜进行物质补偿,再次利用。内吞和循环的平衡维持了质膜物质组成的稳定,保障多种细胞过程正常的进行[3]。


微丝成核促进因子很早被发现存在于内吞体的相关结构,在内吞体形成、成熟和胞内定位等方面发挥作用[4-8]。一般认为,成核促进因子WASH及其激活内吞体上分枝微丝聚合是膜运输的关键调控因素,在retromer介导的内吞体至高尔基体运输中发挥重要作用[7, 9-11]。在循环运输中,微丝结构通常从以下三个方面参与调控:1)底物分选:微丝与循环底物直接结合,使其聚集在内吞体的特定区域上[12, 13]。2)提供机械力的支撑:稳定内吞体的管状结构,创造膜微区域以分隔降解通路[12, 14]。3)聚合产生膜张力:与Dynamin 协同对含底物的循环载体进行膜切割[7, 14, 15]。


为进一步探究循环运输和微丝骨架之间的调控细节,研究人员在线虫中展开全基因组筛选,找寻与循环运输相关的微丝调控蛋白,解析微丝骨架在循环运输的不同步骤中发挥的具体作用。研究人员分别以CIE循环货物hTAC及微丝骨架标记物Utrophin-CH为指标在线虫全基因组中进行筛选,发现了PTRN-1/CAMSAPs、RTKN-1/Rhotekin、WTS-1/LATS等与微丝及循环运输调控相关的候选蛋白。其中,WTS-1/LATS 是在Hippo pathway 中的一种关键激酶,可以结合转录辅激活因子Yki/YAP1,并捕获细胞质中的Yki/YAP1[16]。线虫、果蝇和小鼠wts-1/Wts/LATS的突变体都胚胎致死,证明WTS-1/Wts/LATS在维持动物的生长发育过程中是必需的[17-19]。

线虫WTS-1为哺乳动物LATS的同源物,LATS激酶是Hippo通路中核心组分。研究人员发现wts-1(ycxEx1635)突变导致线虫肠细胞中循环运输货物在胞质中显著累积。同时,在突变体中,微丝架构也受到了严重影响,在胞质中异常聚集。PTRN-1/CAMSAPs促进内吞体上微丝聚合,WTS-1主要定位在分选内吞体上,与PTRN-1具有较好的亚细胞共定位。实验发现,在wts-1(ycxEx1635)遗传背景下,PTRN-1在内吞体上过度累积,提示WTS-1可能抑制PTRN-1在内吞体上的膜定位。PTRN-1缺失可以缓解wts-1(ycxEx1635)中微丝骨架异常聚集表型。此外,UNC-60A/cofilin作为经典的微丝骨架负调控因子,可以介导微丝解聚。LIMK1通过磷酸化cofilin来稳定微丝结构。

之前的研究报道,LATS可以通过抑制LIMK1的活性,使其不能持续磷酸化cofilin,从而促进微丝解聚。在UNC-60A/cofilin(S3A)突变体中,cofilin不再被LIMK1磷酸化,可持续发挥负调控微丝的功能。研究人员发现,在wts-1(ycxEx1635)突变体中,过表达UNC-60A/cofilin(S3A)可使循环运输中的累积表型和微丝聚集表型有所改善,进一步证实了LATS可以正调控cofilin的活性。


综上结果,研究人员提出了一个新的调控模式:WTS-1/LATS通过协同调控PTRN-1内吞体定位和UNC-60A/cofilin的磷酸化,进而抑制微丝结构的过度组装,维持循环运输的有序进行。
