2006年,荷兰阿姆斯特丹大学的Victoria Bock、Henk Hiemstra和Jan van Maarseveen在 European Journal of Organic Chemistry ( EurJOC )上发表了一篇题为“ Cu I -Catalyzed Alkyne–Azide “Click” Cycloadditions from a Mechanistic and Synthetic Perspective ”的小综述论文,针对一个在“点击化学”(click chemistry)领域中最普通的反应——铜催化的“炔—叠氮环加成反应”(alkyne-azide cycloaddition reaction,见下图),对当今这种重要的制备方法进行了总结分析。 Cu I -Catalyzed Alkyne–Azide “Click” Cycloadditions from a Mechanistic and Synthetic Perspective V. Organic Chemistry: A Lack of Challenges?(有机化学,缺乏挑战?) 。 I worry a lot about organic chemistry, about the state of the discipline. I worry about the relative lack of grand challenges, and that most academic work is highly incremental and, worse, almost entirely the result of screening rather than design. There is still so little predictive power (at least in academia) in drug or catalyst discovery. I have a theory that the reason we're so brutal with each other in paper and grant refereeing is because we're essentially dogs under the table fighting for scraps.( 我担心 有机化学 很多,尤其是学科目前的状况 , 我担心 有机化学重大挑战 相对缺乏 ,但是学术论文数量 在迅速 增加 ,可笑的是, 几乎 完全是 在筛选结果, 而并非基于化学原理的设计 。 There are big exceptions, which make me excited to be a scientist. There's usually something in Nature Chemistry that has the wow factor, for example. They're just so rare. . .
