This should be the case if subspecific boundaries are evolutionary faultlines along which speciation is generally more likely to occur. This pattern has been described for birds, but remains poorly understood in mammals. To investigate the relationship between species richness (SR) and subspecies richness (SSR), we calculated the strength of the correlation between the two across all mammals. Mammalian taxonomic richness correlates positively, but only very weakly, between the species and subspecies level, deviating from the pattern found in birds. However, when mammals are separated by environmental substrate, the relationship between generic SR and average SSR in non-terrestrial taxa is stronger than that reported for birds (Kendall'sτ= 0.31, p 0.001). By contrast, the correlation in terrestrial taxa alone weakens compared to that for all mammals (Kendall'sτ= 0.11, p 0.001). A significant interaction between environmental substrate and SR in phylogenetic regressions confirms a role for terrestrial habitats in disrupting otherwise linked dynamics of diversification across the taxonomic hierarchy. Further, models including species range size as a predictor show that range size affects SSR more in terrestrial taxa. Taken together, these results suggest that the dynamics of diversification of terrestrial mammals are more affected by physical barriers or ecological heterogeneity within ranges than those of non-terrestrial mammals, at two evolutionary levels. We discuss the implication of these results for the equivalence of avian and mammalian subspecies, their potential role in speciation and the broader question of the relationship between microevolution and macroevolution.

注:本文已经在“生物探索”网站发表,未经许可,请勿转载。 剑桥大学新研究揭示钠离子通道意想不到的结构 诸平 钠离子(Na + )通道与许多严重的疾病如心脏病、癫痫和疼痛有关系,在研制针对这些疾病的药物时,科学家通常将Na + 通道作为一个重要的潜在目标。但是,对于Na + 通道的具体结构,很多科学家并不熟悉。剑桥大学(University of Cambridge)近期发布了一项新研究,提供了鲜为人知和意想不到的洞察结果——Na + 通道的结构和机制。研究结果发表在《生物化学杂志》(Journal of Biological Chemistry)上。 钠离子(Na + )通道及其主要功能 钠离子(Na + )通道是位于细胞质膜上的一种跨膜糖蛋白,通常由α、β1、β2三个亚单位组成,因其在电刺激的条件下能够被激活开放,故又称其为电压门控钠离子通道,其分布范围非常广泛。钠离子(Na + )通道的开放主要是引起细胞膜外的Na + 内流,改变细胞膜两侧电位的极性,从而造成去极化过程。 Na + 通道中央有能通过Na + 的亲水通道,通道的外端有选择性滤孔,是选择通过离子的部位。孔道中有闸门m门,蛋白质的构象变化使m门开放与关闭。Na + 通道有电压感受器,其上有电荷。这些电荷在电场的作用下移动,造成闸门的开放,因而也记录到门控电流。去极化引起Na + 通道开放后约在2ms内就会失活。从膜内侧施加蛋白水解酶可以阻止通道失活,也就是孔道内端有蛋白质的失活h门。h门一般在m门开放后几毫秒内关闭。 Na + 通道的主要功能是维持细胞兴奋性及其传导。与其他类型的离子通道相比较,Na + 通道研究的比较早,是科学家目前对其特征、分类、结构、门控动力学、生理意义、药理作用等了解比较清楚的一类离子通道。但是,至今Na + 通道的机理与其疾病类型之间仍然尚未完全吻合,而随着对其研究的不断深入,Na + 通道的发病机理与疾病类型的关系将会更加明朗,这将为临床开展预防、诊断以及治疗提供明确的医学基础。 Na + 通道与疼痛的产生有密切的关系 中国科学院上海生命科学院鲍岚研究组的博士研究生张振宁和李乾等,2008年曾经在《细胞科学杂志》( Journal of Cell Science , 2008, 121, 3243-3252. doi: 10.1242/jcs.026856 )发表了他们的研究成果,发现Na + 通道是可兴奋细胞产生动作电位的基础,脊椎动物的钠离子(Na v )通道中的Na v 1.8,是特异性高表达在背根神经节初级感觉小神经元中的一种电压门控钠离子通道,它与疼痛的产生有密切的关系。 Na v 1.8主要驻留在内质网中,其第一个胞内环上的RRR结构域是一个内质网滞留信号,对Na v 1.8驻留在内质网中有贡献,限制了其有效地向细胞膜表面的运输及功能的行使。当Na v 1.8的RRR内质网滞留信号失去功能后,其细胞膜表面表达量较野生型Na v 1.8显著升高。Na v 的β3亚单位通过与Na v 1.8的第一个胞内环结合,掩盖了Na v 1.8的内质网滞留信号,促进Na v 1.8向细胞膜表面的运输。 此项工作首次在Na v 1.8中发现了内质网滞留信号,并对Na v 1.8中内质网滞留信号的功能与调控提供了有力的证据,同时也揭示了Na v β亚单位对α亚单位调控的分子机制,为深入了解疼痛的产生和发展提供了新的研究方向和理论基础。 新研究揭示Na + 通道的结构和新机制 神经和其他电刺激细胞与另外的神经和其它细胞之间通过传输电信号而互相沟通, Na + 通道在这一过程中扮演着至关重要的角色。脊椎动物的钠离子(Na v )通道是由一个离子传导α亚单位和相关的β-亚单位构成。剑桥大学这项新研究的侧重点是β-亚单位中一种称为β3的亚单位。β3位于神经元和心肌细胞中,在Na + 通道调节方面具有特别重要的作用。 在研究中,研究人员使用蛋白质x射线晶体学技术来确定β3-亚单位一部分的原子水平结构——“免疫球蛋白域(immunoglobulin domain)”,并给出其晶体结构。β3-亚单位的该区域位于细胞外,和心脏钠通道α-亚单位结合在一起。研究人员惊讶地发现,3个“β3-免疫球蛋白域”聚在一起形成了一个三聚体。 分析超速离心法证实在游离溶液中,免疫球蛋白域的存在形式有单体、二聚体和三聚体,而原子力显微镜成像结果,也发现了全长的β3亚单位单体、二聚体和三聚体。半胱氨酸残基的突变是维系二聚体和三聚体在三聚体界面不稳定的关键。 随后,研究人员使用原子力显微技术(Dilshan Balasuriya)对单个三聚体进行成像,进一步证明β3亚单位可以与Na v 1.5α亚基上的多个位点结合,诱导α亚单位的寡聚物形成,其中包括三聚体。 这项研究结果显示了Na v 通道中的β3亚单位三聚体与3种钠通道α-亚单位之间的新的和意想不到的交联作用,为一些病理Na v 通道突变提供了新的结构上的深刻理解。 这项研究是在剑桥大学生物系Tony Jackson博士和Dima Chirgadze博士领导之下,主要由Sivakumar Namadurai进行的。Tony Jackson博士说:“我们的结果出人意料,我们一直致力于β3-亚单位研究已经大约有14年了。在此期间,我们不得不间接地在分子水平上进行推断。为了真正看到亚单位的原子结构以及它是如何形成三聚体的,十余年终修正果,犹如突然打开一盏灯泡,罕见的令人惊讶的一幕物瞬间一清二楚地展现在眼前。” Dima Chirgadze博士补充说:“我们的研究对理解钠离子通道机制行为有着重要的意义。迄今为止,一直存在一种假设,即单个钠离子通道的功能是相互独立的。但这种观点可能对于钠离子通道的理解与认识过于简单。一个非常令人兴奋的可能性就是钠离子通道的α-亚单位通过β3-亚单位三聚体交联,可导致一些钠离子通道在功能上是连接在一起的。如果这种观点正确的话,这将允许动作电位一个更有效的启动。” 更多信息请浏览: http://www.biodiscover.com/news/research/110535.html
作者: 追梦九天 (1) Jameson的故事 Jameson 是当今CFD届的超级大牛。偶的超级偶像哦。Jameson是个英国人,出生在军人世家。从小随老爹驻守印度。于是长大了也抗起枪到海外保卫日不落帝国,军衔是Second Lieutenant。无奈“日不落”已落,皇家陆军已经不需要他了。大概有什么立功表现把,退役后就直接进了剑桥大学。在那里拿到博士学位。辗转间从英国来到了美国,从工厂又到了学校。成了Princeton的教授。在那里提出了著名的中心差分格式和有限体积法。就是在这里,发表了他那篇著名的中心差分离散的有限体积法。中心差分格式,大家都知道,是二阶,但是稳定范围特别小,Pe不能超过2,于是就得加人工粘性(一听这名字,数学家就倔嘴巴,不科学嘛),这是大学生都知道的事,怎么加就是学问了。Jameson用二阶项做背景粘性,用四阶项抑制激波振荡(也亏他想得出来),配合他提出的有限体积法,获得了极大的成功,很快风靡世界,工程界几乎无一例外在使用他的方法,原因很简单,他的方法乐百氏,而且又有相当精度。从此大行于市,座上了P大的航空系系主任,也确立了CFD界第一大牛人的地位。 Jameson发文章有个特点,喜欢发在小会议上或者烂杂志上,反正是SCI检索不到地方。包括后来关于非结构网格,多重网格等等经典的开创性文章,都是这样。(如果按照清华的唯SCI论的评判标准,我估计在清华最多只能给他评一个副教授当当。)牛牛的人总是遭人忌妒,哪里都这样。看着Jameson的有限体积方法这么受欢迎,有些人就红眼了。于是说,有限体积方法不错,可惜只适合于定常问题计算,非定常计算就不怎么样嘛。 Jameson那里能容忍别人对他的得意之做胡说。于是,灵机一动,想出了一个双时间尺度的方法,引进一个非物理时间,把非定常问题变成了一个定常问题计算,还真好使,又风靡世界,从此天下太平。97年,Jameson年龄到了,就从P大退休了,结果又被聘请到Standford大学当Thomas V. Jones Professor搞起了湍流来。前不久偶导师见他回来,对欧们边摇头边说,“几年不见,老得快不行了”,言下之意,我们如果想多活几年,不要去搞什么湍流。 (2) Steven A. Orszag的故事 Steven A. Orszag是一个天才级别的人物啦。在直接数值模拟,谱方法,湍流模型等等许多方面都有开创性的贡献。天才嘛,总是有缺陷的,不是生活不能自理,就是不懂得处理人际关系。前者还好办,只是lp不舒服,后者嘛,让同事和同行不舒服,可麻烦就大了。不幸的是,Orszag属于后者。对于他的恃才傲物,有人早就恨得牙根痒痒,报复的机会终于来了。 三十年前,湍流模型的先驱们,是通过数值试验,再连懵带猜的确定下了双方程湍流模型的参数。20年前,Orszag突发奇想,能否用RNG(重整化群理论)从理论上推导这些参数呢?RNG理论在相变上取得了很大的成功,发明者也在81年获得了 Nobel奖。牛人就是牛人很快居然真从理论上推出了这些参数。这下湍流模型界可炸开了锅,这岂不是要砸掉很多人的饭碗?这不等于说那些老家伙几十年前的工作一钱不值么?这帮大学霸可不是省油的灯。环顾地球之大,Orszag居然找不到一本杂志愿意接受他这篇文章。Orszag这个郁闷呀,这个生气呀,好歹庵也是绝代高手嘛,昨这么不给面子呢?他一气之下干脆自己扛杆旗,办份杂志,自己当主编,自己出版,看谁说闲话。1986年,《Journal of Scientific Computing》终于开张了。第一篇文章就是“Renormalization Group nalysis of Turbulence: I Basic Theory”。这篇文章很快获得了大家的广泛认同。但是对RNG的攻击并没有到此为止。偶看到最搞笑的是一个牛牛(不想提他的名字了)在AIAA J. 上的一篇文章。当然是吹自己的模型计算比标准双方程模型多么多么的好。都已经比较结束了,他还觉得不过瘾,话锋一转,把RNG模型胡算一把,然后一桶狂批,还煞有介事的分析为啥算不好。其实我倒觉得,既然RNG能够从理论上推导出他们当年胡乱搞出来的参数,不正是对他们工作的证明么?能够从完全黑暗的世界寻找到这些参数,这除了天才,还能说什么呢? (3) Godunov的故事 Godunov大家都晓得吧,迎风类型格式的开山鼻祖。二十世纪CFD的数值方法基本上是沿着他老人家开创的Godunov类型格式的方向发展。连如今大姥级的Roe,van Leer都要发文章pmp,毕竟他们都是靠着老大发家的嘛。他座上老大宝座的屠龙刀-Godunov格式,实际上是1954年他25岁时候的博士论文。老板上课时候曾经讲,当时不知道为啥他得罪了苏维埃政府要砍他的头,于是他一着急,弄出了这把屠龙宝刀,拣回了小命(不过这个传闻,我没有找到相关的文献得以证实,好在我相信偶老板读的书比我多,二来嘛本来就是八卦系列也无所谓了)。 我现在就来讲讲有根有据的东西,老大是怎么弄出这把屠龙刀的。1954年春天,苏联的第一台电子计算机“Strela”就将送到老大当时所在的单位Keldish Institute of athematics,上级要求他们弄几个格式来算一算。当时一个叫Zhukov的人就弄出了一个东西。这家伙也算是个牛人了,弄出来的这个东西,同1年后 P.D Lax的CFD奠基性名著中提出的东西是完全一样的。可惜呢,这家伙数学不好,他是连蒙带猜弄出来的,尤其是为了自圆其说的那几个假设,现在回过头来看根本就是错误的,是推不出这个结果的。当时为了弥合这个问题,就请来了Godunov看能不能解决这个问题。结果一发不可收拾,居然就借此搞出了 Godunov格式。后来老大回忆刀,幸好当时他没有看到Lax的文章,要是看了,压根就不会有Godunov格式了。(If I would have read Lax’s paper a year earlier, “Godunov’s Scheme” would never have been created.)这么重大的贡献得发文章让大家都晓得才行呀。老大于是一毕业就四处投杂志,他先投了一家叫Applied Mathematics and Mechanics的杂志,杂志居然把他据了,理由是,老大的工作是一个纯粹的数学工作,没有做任何关于力学的研究。老大一想也对,他本来就是数学家嘛,于是他改投一个纯数学的杂志,谁知道,没过多久,又被退稿了,这次的理由是,老大的工作是一个纯力学的研究,没有任何关于数学的内容。老大当场晕倒。后来老大又投了几家还是不中,这下没有办法了,老大只好找后门,托他的老板Petrovskii了,正好老板是Mathematicheskii Sbornik杂志的编辑,终于在1959年,毕业四年后这篇文章发表在了这个杂志。

加入执政党的党员,原因不易单一化地叙述。 加入还在奋斗期间的党、或永远不能执政的党,其党员绝大多数是为了理念。 英国共产党(1920-1991)恐怕从来没有执政的机会。英共党员多彩多姿,如结构生物学先驱(被人称Sage/智者的)JD Bernal(他的学生诺贝尔奖得主Hodgkin不是党员,但她丈夫曾是)、著名遗传学家JBS Haldane、进化论学者John Maynard Smith,其中不乏有剑桥大学、牛津大学的教授。他们入党,对自己的学术生涯有影响,可以说基本是负面的。最简单的是,号称无政治歧视的美国,禁止共产党员入境,无论党员是英国剑桥大学教授、是世界著名科学家、还是是美国需要征求特定领域意见的皇家学会会员,都被这样对待。 英国著名科学家Anne McLaren(1927-2007)就曾经几十年被美国拒之门外。 Anne McLaren出生于英国贵族家庭。这个富有的家庭政治却不保守右倾,而偏自由主义。她9岁时作为童星拍过电影。她在牛津大学念动物学,曾跟随JBS Haldane研究果蝇。博士期间研究遗传和病毒。1952年,25岁获博士学位,并与同学Donald Michie结婚。其后他们在伦敦合作到1959年。他们都信奉产生在他们国土的马克思主义、加入了英国共产党,即使在1956年因为苏联入侵匈牙利而导致英共党员数量减少后,他们还是党员。 Michie是英共机关报《每日工人报》(The Daily Worker)的记者、McLaren曾任牛津的英共支部书记(她的支部曾有一百多位党员)。美国拒绝McLaren的入境一直持续到1991年,那时英国共产党也解体了,而她已经做皇家学会会员十六年。如果美国继续拒绝她入境,可能要和英国科学交往出问题:McLaren于1991年至1996年任皇家学会外事秘书和副会长,成为皇家学会历史上第一位女性“官员”。1993年她因为自己的科学成就而册封DBE。1959年她和Michie离婚后,他们保持良好关系。2006年,Michie的妻子Jean被诊断患癌症后,他们搬到McLaren处合住。Jean于2006年过世后,McLaren和Michie买了一栋三层的楼房,她们分别住两层,另外一层接待孩子。2007年,McLaren和Michie一道因车祸丧生。 她自上大学以后爱科学,但认为科学工作者应该照顾家庭。看到有孩子的科学家5点半还没回家,她说你现在该回家和孩子在一起了。 她对学生很好,她也乐于助人。中国河南科技学院的刘用生老师在写文章时,素昧平生的McLaren不仅给他多次修改文章,还特意翻译一篇俄文文献,给刘老师做参考。她支持中国人,不是政治原因,而是因为她热爱科学和热情支持其他人。有个相关的题外话:结构生物学家Dorothy Hodgkin,1970年代也曾支持中国做胰岛素结构的科学家,先是教过中国为此派遣的留英学者、后是在国际上推崇北京解析胰岛素晶体结构的研究工作,肯定北京结构的分辨率比自己的高。因为Hodgkin从1930年代就做胰岛素结晶,到后来还在做,所以,看起来是为自己的“竞争对手”说好话,如果她和其他很多人一样看待科学研究的话。 McLaren一贯保持左倾政治态度,为社会正义努力,捍卫弱者权力。早年也在系里面参加左派右派辩论。当年参加世界和平大会,后来多次参加反战游行。在西方宗教和右倾反对研究人类干细胞时,她以科学家身份支持研究人类干细胞。 我最初注意McLaren的研究,是在写决定男性的基因科普文章时。哺乳类、包括人类,雌雄是怎么决定的?这是有趣而重要的科学问题。发现决定哺乳类性别的基因,主要依赖于研究人类一些变异个体和老鼠。流行了十年的一个假设是H-Y可能是决定男性的分子。McLaren在1984年发表论文,排除了H-Y参与男性决定的可能。这个领域积极再寻找其他可能的基因,经过麻省理工的David Page等的努力,最后于1990年,澳大利亚的Marshall Graves 和英国的Lovell-Badge两实验室发现了决定雄性的基因SRY。 McLaren在五十年代研究过如何使老鼠的早期胚胎可以在体外培养,她们摸索条件,使老鼠胚胎在体外培养以后,可以再回到子宫内发育成为个体。这部分工作,有助于探索人类体外受精的技术。她的科学兴趣集中于种系发生细胞(卵细胞和精细胞),她认为它们是最有趣的细胞。她从大学开始从事科学研究,直到2007年80岁去世。 Anne McLaren的研究论文,2008、2009年还在发表。 因为入党原因不同,有些加入执政党的党员,可能不一定做得到预期的为公众利益、为公平正义,而为自己利益和逃避责任的也不能算很少。即使在教育和科研机构,恐怕能做到McLaren这样程度的不多,有些可能还是反面。而虽然McLaren所属的党于1991年以后不复存在,她的行为却更像有些人们的期望。 为了理念而入党的人,即使党没了,他们也会坚持自己的信念、做自己认同的事情。他们得到的尊敬,不限于和他们在政治上理念和意见相同的人。 McLaren A, Biggers JD (1958). But all her behavior smelled of petit-bourgeois. It seems to me that her greatest desire in life was to raise herself up to a higher stratum. Every time she set her aspiration at a target not too high to attain; bue as soon as it is reached, at once she would set another a little higher. She never felt satisfied. Besides fulfilling her aspiration , there was nothing else she cared. At first she just wanted to lift herself out of her poverty-stricken family. Repressing the ideal that she should have a house equipped with beautiful furniture of her own besides getting a husband, she jumped at the chance of marrying Bob soon after their acquaitance. As a consequence of her mother-in-law’s death, she started to manage the house. One afternoon, when she was window shopping in a furniture store, her dream of refurbishing the house vevived. With a yielding and uxoriors huaband, her greed to grab what was there for her knew no limit. After nursing a period that taxed patience, she satisfied herself with selling the old furniture and replacing them with fashionable ones. Following the success, she managed to urge her husband to see Maud, intending to throw Old Thompson over to her. While he was away running her errand, she sold away the old man’s tall-backed chair too. Among a chain of events, these two were the last straw for Old Thompson. He uttered his wrath to his son and daughter-in-law: “I am waiting, just bidding my time till the good Lord decides to take me to her again. And you’ll just have to bide your time and wait for me." I think the reason why readers are more often than not disgusted with Annie is that she just could not fobear to wait. Her eagerness to get things done is in itself blameless. (To digress a little, many of us who are faced with the task of accelerating “four modernizations” lack this kind of eagerness.) Fortunately, Annie was not so base as to be devoid of all conscience. She did regret that she had sold the chair and her repentance would result in a better treatment of her father-in-law afterwards. After all, she should have realised that the world was not creted for her alone.

Her focus is on a network of three interacting hormones that move over long distances in the plant, providing a rich source of systemic information that is locally interpreted to regulate branching. She is interested in the biochemical and physiological modes of action of these hormones and how they relate to whole plant responses. Central to her approach is the combination of genetic analysis in the model plant Arabidopsis, and computational modelling of the system.
