肌肉与代谢——“施瓦辛格小鼠” 最近刚卸任的州长施瓦辛格,闹出不少的绯闻。做州长的施瓦辛格并没有给众人留下显赫的政绩,到是他强壮的肌肉及绯闻会给人留下不少的话题。 但你们想到过吗?在实验室里,小老鼠也可以变身鼠中“施瓦辛格”?依赖于肌肉蛋白质合成。在骨骼肌蛋白质代谢中,促进蛋白质的激素包括睾酮、胰岛素,以及相关生长因子胰岛素生长因子(IGF-1)及异构体机械生长因子(MGF)等。还有抑制肌肉生长的肌肉抑制素(myostatin)等。在肌肉的生长及壮大方面维持平衡。通常抗阻力量练习及优质蛋白质补充有助于肌肉壮大,肌肉尤其对机械运动刺激更敏感。利用特定的遗传差异,科学家创造了小鼠中的"施瓦辛格",它的肌肉质量超过普通小鼠四倍。该研究成果有助于探索人类肌肉萎缩症、艾滋病以及癌症引起的肌肉损失。通过转基因技术,一方面可以通过变异—让它们不能制造肌肉抑制素(myostatin),但却能大量生产卵泡抑制蛋白(follistatin)。由于肌肉抑制素会限制肌肉发育,因此缺乏它的小鼠会多长肌肉;同时,卵泡抑制蛋白会影响抑制肌肉发育的蛋白起作用,也就是说,两方面因素都促使小鼠的肌肉更多更强壮,该方面已经在养殖业应用,如动物的产肉量增加。还可以通过IGF-1基因改造,让肌肉一块块地隆起,或外源性补充类固醇激素,曾被一些力量项目的运动员采用。 通过这类技术制造的“施瓦辛格鼠”与正常鼠的肌纤维存在很大不同,前者肌纤维的大小超过后者的两倍,多117%,而且在数量上也比后者多73%。研究人员认为,在缺乏肌肉抑制素的条件下卵泡抑制蛋白仍然能够促进肌肉发育,这说明小鼠体内存在不止一种肌肉调控机制。如果生物学家能够在人体内确定更多的卵泡抑制蛋白可以绑定的蛋白,就有望导致新的促进人类肌肉发育或者维持肌肉的药物出现。此外,新的研究成果的意义还在于培育更加长肉的家畜。不过,李塞金特别强调,他不希望新的研究成果成为运动员提升肌肉水平的工具。如果这类被大量应用,可能对整个生物界是一种灾难,但愿仅停留在实验室阶段。
