世界上的生物体个头大小千差万别,甚至亲缘关系很近的物种间,也有成百上千倍的差别。大家最熟悉的是猫和老虎,二者长得很像,属于同一个科,猫科,体重差别上百倍。几年前,Science报道了像水牛一样大的老鼠( A Rodent as Big as a Buffalo )。这些个头差异是怎样进化的呢?看过一些恐龙记录片的网友大概知道,恐龙时代,哺乳动物的祖先刚出现在地球上时,大小就像现在的老鼠。恐龙灭绝后,哺乳动物的个头猛增,出现了像猛犸象、鲸鱼之类的巨兽。 最近PNAS发表的一项研究显示,陆地动物体重增加一千倍大约需要5百万代,水生动物(如鲸鱼)快一些,需要大约3百万代。更有趣的是,作者发现,个体变小得进化就快多了,比变大相同倍数所需的时间快了10倍以上。也就是说,大动物中进化产生小动物很容易,但小动物中进化产生大动物就难了。 为什么变小容易、变大难呢?由于无法访问全文,暂时不知道作者有没有深入讨论其中的原因。我做一点猜测,大家一起讨论。 进化快慢可能的决定因素存在于生物进化的三个基本方面:一是基因突变,二是自然选择,三是随机漂变。 会不会个体大的突变发生频率远低于个体变小的突变呢?我认为这种可能性不大。一些影响个体大小关键基因,活性增强突变会使个体变大,活性减弱突变会使个体变小。这一问题,我们可以从癌症研究中获得一些启示。癌症的发生,既有原癌基因活性增强的突变引起的,也有肿瘤抑制基因活性减弱的突变引起的。也就是说,现有的正常基因,活性变强和活性变弱的突变都很常见,虽不敢说绝对相同,至少可以说差别不太大。 另一种可能就是,变大和变小的突变虽然在自然界中以类似的频率出现,但变大的突变比变小的突变适应环境的可能性小。大的容易被淘汰,小的容易活下来。想想自然界养活一种大象需要一个什么样的生境(或专业一些,生态位),而养活一种老鼠又需要多少空间和资源。变大的突变使自己进化的路越来越窄,变小的突变使自己进化道路越来越宽。小个子们高兴一下吧,我们有进化优势(开个玩笑)。 最后一种可能,随即漂变。生物界存在这样一个规律,生物体个体越大,有效种群越小,自然选择的效率越低,随机因素对进化的影响越大(这里面的背景知识,请查阅种群遗传学有关书籍)。使个体进一步变大的突变,即使有利于个体的生存和繁衍,也不一定能在自然选择中保留下来。相反,个体变小,随机因素的作用变弱,只要变小的突变有利就会很快在种群中扩散。但我认为这种可能性不大。感觉新变异对现有种群大小的影响不会太大。 参考文献 Richards S, There and Back Again. http://the-scientist.com/2012/02/01/there-and-back-again/ 。 Evans AR et al. 一对鹤发童颜的老人,簇拥在满堂的儿孙中间,其乐融融。这是中国传统的幸福观念,国人几千年来的追求。 然而现实与梦想总是保持很大距离。现代人相对于祖辈们,可以说健康长寿了,但儿孙满堂的幸福却没有了,取而代之的是孤独的晚年。 最新研究发现,未开化的动物们也是如此。健康长寿和儿孙满堂之间只能二选一。 细胞内有一类蛋白质,叫分子伴侣(molecular chaperones)。这些蛋白质多才多艺。它们的基本功能是辅助蛋白质折叠。但同时,它们参与细胞对抗冷、热、DNA损伤等各种不良刺激。如果细胞中分子伴侣表达水平高,也就是这些蛋白质多一些,细胞就“身强力壮”、不怕各种小灾小病。而分子伴侣少的细胞,那就是弱不经风、整天病病歪歪了。 那就提高分子伴侣的表达量吧。研究发现,提高分子伴侣的表达量确实可以使线虫健康长寿。但同时,分子伴侣的增加降低了线虫的繁育能力。在不良环境中,分子伴侣多的个体,身强力壮,是一大优势。但如果环境条件优越,分子伴侣少的家族才会人丁兴旺。自然界没有永远的温柔乡、也没有永远的逆境。环境不断变化,不同特征的生物体也就十年河东、十年河西。 我猜测,人类、大象等动物,长寿的进化过程中可能就是以降低生育率为代价。 参考文献: Ruth Williams (2011) The Tradeoff of Stress. http://the-scientist.com/2011/12/15/the-tradeoff-of-stress/ M. 定向选择 定向选择( directional selection ):寄主抗病品种有利于病原物相应的毒力基因的选择作用,称为定向选择。定向选择 (Directional selection) 是天择的一种形式。 对于一种特征 ( 性状 ) 的不同状态而言,因为不同状态带给生物体的适应度 ( 适存度 , fitness) 有差异,其中一种状态的适应度较高,具有此状态的有利个体就能产生较多的子代,造成此状态的等位基因 (allele) 便倾向于在族群中增加,并渐渐取代其他等位基因,此过程即称为定向选择。 假设,影响一特征 / 性状的等位基因有 A1 与 A2 两种,如果同型合子 A1A1 的适应度最高,而异型合子 A1A2 适应度中等, A2A2 适应度最低(ω 11 ω 12 ω 22, ω代表适应度),在天择的影响下,保留 A1 、并淘汰 A2 的过程就是定向选择。若环境维持稳定, A1 基因所代表的性状持续带给此物种较高的适应度,“有利的” A1 基因随着时间而渐渐散布到整个族群,基因频率提高,最后达到稳固 (fixation) 。 接续上例,适应度较低的 A2 基因可能会完全消失于族群中,此过程可称为净化选择 (purifying selection) 。对于一个突变的新基因型,而受到定向选择而被保留,可称为正向选择;反之,有害的基因受到淘汰称为负向选择。依照讨论的例子,有时负向选择与净化选择意义相同。 定向选择与平衡选择 (balancing selection) 相反,前者只保留有利的基因型,后者会倾向于保留多种基因型。值得考虑的是,不同的环境可能偏好不同的基因型,因此一个等位基因有利或有害,需要参考生物族群的生活环境才能决定。 定向选择造成的基因频率变化,由相对的适应度决定,并不会因为此基因是显性或隐性而受影响。虽然说,被保留的基因达到稳固的时间,会因为此基因是显性或隐性而有差异:若有利的基因是隐性,则在起始基因频率很低的状况下,只有很少的个体是隐性同型合子、很少个体表现出较高的适应度,因此需要累积很多世代才能增加基因频率。 以族群遗传学的方法,比较两世代之间的基因频率是否有变化,可以推知此基因所影响的性状是否正在受到天择的作用。 自然选择 正负中性选择 自然选择 Natural selection 生物在生存斗争中适者生存、不适者被淘汰的现象。最初由 C · R ·达尔文提出。达尔文虽借用了人工选择中的“选择”这个词,但这只是一种比喻,并非说有超自然的力量在进行选择。按照达尔文的意见,自然选择不过是生物与自然环境相互作用的结果。从进化的观点看,能生存下来的个体不一定就是最适者,只有生存下来并留下众多后代的个体才是最适者;又考虑到进化是群体而不是个体的现象,现代综合进化论从群体遗传学的角度修正了达尔文的看法,认为自然选择是群体中“不同基因型的有差异的(区分性的)延续”,是群体中增加了适应性较强的基因型频率的过程。 自然选择是为了使物种更好的适应环境,另一方面是为了抑制生物过度繁殖生物在生存过程中,既要与自然环境进行斗争,又要与其他生物进行斗争。在生存斗争中,具有有利变异的个体容易生存下来(适者生存),具有不利变异的个体则被淘汰,这便是自然选择的原因,自然选择促成了物种的进化。 达尔文的自然选择学说,其主要内容有四点:过度繁殖,生存斗争(也叫生存竞争),遗传和变异,适者生存。 正向选择,负向选择(纯化选择)和中性选择 Positive selection , Negative ( Purifying ) selection and Neutral selection In population genetics, the relative fitness of a new mutant allele a relative to the predominant wild-type allele A is measured by the selective coefficient (Malthusian parameter) s. Then s 0, = 0, 0 correspond to negative (purifying) selection, neutral evolution, and positive selection, respectively. The frequency of the new mutant allele goes up or down over generations, affected by natural selection as well as random genetic drift. Whether random drift or selection dominates the fate of the mutation depends on Ns, where N is the effective population size.
