研究人员从32个正常人体组织内超过12,000个基因中定量了相对蛋白质水平。组织特异性或组织富集的蛋白质被鉴定,并将其与转录组数据进行比较。许多普遍的转录本被发现编码组织特异性蛋白质。RNA和蛋白质富集的不一致揭示了分泌蛋白质合成和工作的潜在位置。蛋白质的组织特异性分布还提供了对复杂生物学事件的深入视角,这些事件需要多个组织的相互作用。

Abstract: Determining protein levels in each tissue and how they compare with RNA levels isimportant for understanding human biology and disease as well as regulatory processesthat control protein levels. We quantified the relative protein levels from over 12,000genes across 32 normal human tissues. Tissue-specific or tissue-enriched proteinswere identified and compared to transcriptome data. Many ubiquitous transcripts arefound to encode tissue-specific proteins. Discordance of RNA and protein enrichmentrevealed potential sites of synthesis and action of secreted proteins. The tissue-specificdistribution of proteins also provides an in-depth view of complex biological eventsthat require the interplay of multiple tissues. Most importantly, our study demonstratedthat protein tissue-enrichment information can explain phenotypes of genetic diseases,which cannot be obtained by transcript information alone. Overall, our results demonstratehow understanding protein levels can provide insights into regulation, secretome,metabolism, and human diseases.
